Abstract

Polymeric microparticles were prepared by an O/W- solvent evaporation method, whereby an organic polymer solution was emulsified into an external aqueous phase followed by solvent evaporation. The particle size/distribution of the emulsion droplets/hardened microparticles was followed by FBRM by monitoring the chord length distribution (CLD), particle size mean and chord count of microparticles. Various formulation parameters including polymer solution concentration, volume of external aqueous phase, stirring speed and combinations of two microparticle formulations (combinations of different stirring speeds) emulsified in the same external phase were investigated. The morphology of EC microparticles was characterized by optical microscopy and scanning electron microscopy (SEM). The solidification time of the emulsion droplets into solid microparticles could be determined via FBRM and occurred within 20 minutes with the solidification time being more rapid at higher stirring speeds and with solvents of higher water solubility. Emulsification of the second polymer formulation after hardening of the first polymer formulation resulted in the formation of a blend of two separate microparticle formulation in one process when compared to two separate preparation processes followed by blending of the individually prepared microparticle formulations.
