Abstract

This study was carried out at the Plant Biotechnology Laboratory, Agricultural Faculty, University of Jambi from February through to September 2006 with the aim of obtaining the concentration of putrescine that was effective for embryogenesis induction on soybean anther culture. The investigation consisted of various concentrations of putrescine (0, 2,5, 5,0, 7,5 and 10,0 mM) that were tested on media with two different composition of plant growth regulators, i.e. 20 M 2,4-D + 10 M kinetin and 20 M 2,4-D + 20 M kinetin. Anthers were obtained from floral buds sizing 2,5 – 3,5 mm from soybean cultivar Merubetiri (results of first and second year of study), cultured on solid MS medium supplemented with vitamins and 30% sucrose. Cultures were placed on racks in culture room with 16-h photoperiod and light intensity of 50 µmol m-2 sec-1. Temperature was set at 24 – 26 oC. Each trial consisted of 5 replicates, and each replicate consisted of one culture flasks with 10 anthers obtained from the same floral bud. Response showed by the anthers of soybean cv. Merubetiri cultured on solid MS medium supplemented with various concentration of putrescine and 20 M 2,4-D + 10 M kinetin or 20 M 2,4-D + 20 M kinetin, was in the form of callus proliferation within 4 to 20 days following culture initiation. Callus proliferation was preceded by a swollen on the surface of anthers, followed by changing in color from light green to brownish. Following this, anther wall turned into amorphous shape, before finally was covered by a white, creamy, brownish, light green and/or dark green callus mass. Initially, the callus showed friable or compact structure, but following two weeks of proliferation the majority of callus showed compact structure with embryogenic callus properties. Among putrescine levels tested, the best callus proliferation occurred on the concentration of 2,5 M; when the concentration of putrescine exceeded 2,5 M, callus proliferation will be significantly inhibited.
