Abstract
RINGKASAN Kopi arabika merupakan salah satu jenis kopi yang banyak dikonsumsi hampir sebagian besar penduduk didunia karena kopi tidak hanya memberikan aroma dan rasa yang khas tetapi juga memberikan banyak manfaat bagi kesehatan. Biji kopi arabika dapat memberi banyak manfaat bagi kesehatan karena mengandung senyawa golongan alkaloid, flavonoid, saponin, fenolik, steroid dan tanin dengan berbagai bioaktivitas seperti antioksidan, antibiotik, dan antiinflamasi. Biji kopi arabika mengandung senyawa bioaktif alkaloid, flavonoid, tanin dan saponin yang dapat berperan sebagai antibakteri. Sehingga penelitian dilakukan isolasi senyawa metabolit sekunder fraksi etil asetat biji kopi arabika dan uji aktivitas antibakteri terhadap bakteri S. epidermidis dan E. coli. Isolasi senyawa metabolit sekunder fraksi etil asetat dilakukan pemisahan menggunakan Kromatografi Vakum Cair (KVC) dan dilanjutkan pemisahan dengan Kromatografi Kolom Gravitasi (KKG). Isolat yang diperoleh dilakukan karakterisasi menggunakan instrumen spektrofotometer UV-Vis dan FT-IR, serta dilakukan penggujian aktivitas antibakteri menggunakan metode kertas cakram. Pada proses pemisahan menggunakan KVC dengan fase diam yaitu silika gel dan fasa gerak pelarut dengan kepolaran bertingkat yaitu n-heksan : etil asetat dan etil asetat : metanol. Diperoleh 26 vial yang selanjutnya dikelompokkan berdasarkan pola noda hasil Kromatografi Lapis Tipis (KLT) didapat 3 fraksi. Pada fraksi 2 dilakukan Kromatografi Kolom Gravitasi (KKG) didapatkan 5 fraksi. Proses isolasi ini menghasilkan isolat pada Fraksi 2.3, isolat yang diperoleh berupa padatan putih dan jika dilihat melalui karakretrisasi termasuk senyawa alkaloid yang diduga senyawa kafein. Hasil uji aktivitas antibakteri terhadap bakteri S. epidermidis dan E. coli menunjukkan isolat etil asetat memiliki aktivitas terhadap bakteri S. epidermidis dan tidak memiliki aktivitas terhadap bakteri E. coli. SUMMARY Coffee arabica is one type of coffee that is consumed by most of the world's population because coffee not only provides a distinctive aroma and taste but also provides many health benefits. Arabica coffee beans can provide many health benefits because they contain alkaloids, flavonoids, saponins, phenolic compounds, steroids and tannins with various bioactivities such as antioxidants, antibiotics, and anti-inflammatory. Coffee arabica beans contain bioactive compounds of alkaloids, flavonoids, tannins and saponins that can act as antibacterial. Therefore, the research was conducted to isolate secondary metabolites of the ethyl acetate fraction of Coffee arabica beans and test its antibacterial activity against S. epidermidis and E. coli bacteria. Isolation of secondary metabolite compounds from the ethyl acetate fraction was separated using Vacuum Liquid Chromatography (KVC) and continued with Gravity Column Chromatography (KKG). The isolates obtained were characterized using UV-Vis and FT-IR spectrophotometer instruments, and antibacterial activity was tested using the paper disc method. In the separation process using KVC with a stationary phase, namely silica gel and a solvent mobile phase with graded polarity, namely n-hexane: ethyl acetate and ethyl acetate: methanol. There were 26 vials which were then grouped based on the stain pattern as the result of Thin Layer Chromatography (TLC) obtained 3 fractions. In fraction 2, Gravity Column Chromatography (KKG) was performed to obtain 5 fractions. This isolation process resulted in isolates in Fraction 2.3, the isolates obtained were in the form of white solids and when viewed through characterization, they included alkaloid compounds which were suspected to be caffeine compounds. The results of the antibacterial activity test against S. epidermidis and E. coli showed that the ethyl acetate isolate had activity against S. epidermidis bacteria and no activity against E. coli bacteria.
