Abstract

Singkong (Manihot esculenta Crantz) merupakan salah satu tumbuhan yang banyak diproduksi di Indonesia. Dimana daun singkong sudah banyak digunakan sebagai pengobatan dalam masyarakat. Daun singkong berkhasiat sebagai antioksidan, antimikroba, antiinflamasi. Daun singkong menunjukkan kandungan senyawa alkaloid, flavonoid, tannin, fenolik, dan saponin. Karakterisasi sebagai tahap awal untuk mengetahui mutu ekstrak, agar ekstrak yang diperoleh aman digunakan sebagai bahan obat. Karakterisasi meliputi karakterisasi spesifik dan non spesifik. Metode. Karakterisasi dilakukan dengan menetapkan parameter spesifik dan nonspesifik dari ekstrak daun singkong, selain itu dilakukan juga skrining fitokimia pada ekstrak. Hasil. Hasil pengujian parameter spesifik karakterisasi ekstrak daun singkong menunjukkan identitas ekstrak yang digunakan yaitu ekstrak air daun singkong (Manihot esculenta Crantz), dengan organoleptik ekstrak berwarna coklat kehita�man berbau khas singkong dan rasa pahit keasaman, kadar senyawa larut air 37%± 4. Hasil pengujian parameter nonspesifik karakterisasi ekstrak air daun singkong menunjukkan kadar abu total 2,33%±0,577, kadar abu tidak larut asam 0,67%±0,288, susut pengeringan 1,395%± 0,040, kadar air 0,748%±0,100, bobot jenis 0,921gr/ml±0,025, cemaran mikroba 7910 cfu/ml, cemaran kapang 6476 cfu/ml, hasil uji kualitatif cemaran logam menunjukkan hasil negatif (Hg, Pb, Cu, Cd). Dan untuk semua hasil hasil pengujian masih berada pada batas normal yang telah ditentukan. Dari hasil penelitian yang dilakukan secara skrining fitokimia dan Kromatografi Lapis Tipis dapat disimpulkan bahwa ekstrak daun singkong (Manihot esculenta Crantz) mengandung senyawa flavonoid, tanin, alkaloid dan fenol. Cassava (Manihot esculenta Crantz) is one of the most widely produced plants in Indonesia. Where cassava leaves have been widely used as a treatment in society. Cassava leaves are efficacious as antioxidants, antimicrobial, anti-inflammatory. Cassava leaves contain alkaloids, flavonoids, tannins, phenolics, and saponins. Characterization is the first step to determine the quality of the extract, so that the obtained extract is safe to use as a medicinal ingredient. Characterization includes specific and non-specific characterization. Method. Characterization is done by setting specific and non-specific parameters of cassava leaf extract, besides that, phytochemical screening is also carried out on the extract. The result. The results of testing the specific parameters of characterization of cassava leaf extract indicate the identity of the extract used, namely water extract of cassava leaves (Manihot esculenta Crantz), with organoleptic extracts blackish brown with a characteristic smell of cassava and a bitter taste of acidity, the content of water soluble compounds is 37%± 4. The results of non-specific parameter testing of characterization of cassava leaf aqueous extract showed a total ash content of 2.33%±0.577, acid insoluble ash content 0.67%±0.288, drying loss 1.395%±0.040, water content 0.748%±0.100, specific gravity 0.921 gr/ml±0.025, microbial contamination 7910 cfu/ml, mold contamination 6476 cfu/ml, the results of the qualitative test of metal contamination showed negative results (Hg, Pb, Cu, Cd). And for all test results are still within the normal limits that have been determined. From the results of research conducted by phytochemical screening and Thin Layer Chromatography, it can be concluded that cassava leaf extract (Manihot esculenta Crantz) contains flavonoid compounds, tannins, alkaloids and phenols.
