Abstract
This study was aimed at investigating the growth and development of potato shoot explants as affected by dark various periods (0, 4, 7 and 10 days) at culture initiation, followed by culturing onto solid MS medium supple¬mented with 0.5 µM IAA plus kinetin at various concentrations (0.1, 1.0, 2.0, 10 and 20 µM). Explants were ob¬tained from adventitious shoot at approximately 5 cm long taken from tubers of potato cv. Granola obtained from Balai Benih Induk Kentang, Desa Batang Sangir, Kabupaten Kerinci, Jambi. Cultures were maintained at 25 ± 1 oC with light intensity of 50 µmol m-2 s-1 and photoperiod 16 hours, except those treated with dark pe¬riod. The results showed that culture incubation at total dark condition for 7 days produced better shoot and root growth than other treatments. At 10 days incubation, all cultured explants died. In addition, 0.5 µM IAA + 0.5 µM kinetin produced the best shoot and root growth during sub-culture. All explants died when cultured on medium supplemented with 0.5 µM IAA plus kinetin at 2.0 µM or more.
