Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh antibakteri spray hand sanitizer dari ekstrak daun buta-buta terhadap pertumbuhan S. aureus dan untuk mengetahui konsentrasi optimal dari ekstrak daun buta-buta sebagai antibakteri spray hand sanitizer terhadap bakteri pertumbuhan S. aureus. Penelitian ini dilakukan di Laboratorium Pendidikan Biologi Universitas Jambi dan UPT Laboratorium Dasar dan Terpadu (UPT-LDT) Universitas Jambi pada bulan Februari sampai dengan Maret 2022. Penelitian ini merupakan penelitian eksperimen, rancangan penelitian yang digunakan adalah metode rancangan acak lengkap (RAL) dengan 5 perlakuan dan 5 pengulangan. Perlakuan meliputi pemberian hand sanitizer terhadap bakteri S. aureus yaitu kontrol (hand sanitizer komersial), hand sanitizer ekstrak daun buta-buta dengan konsentrasi 10%, 30%, 50%, dan 70%. Parameter penelitian yang diamati meliputi zona hambat, pH, dan uji kecepatan mengering hand sanitizer. Analisis data menggunakan One Way Analysis of Variance (ANOVA) dan apabila terdapat pengaruh perlakuan maka dilanjutkan dengan uji Duncan’s New Multiple Range Test (DNMRT) pada taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa hand sanitizer ekstrak daun buta-buta berpengaruh terhadap pertumbuhan bakteri S. aureus dengan rata-rata diameter kontrol (hand sanitizer komersial) (P0) 19,09 mm, (P1) 14,50 mm, (P2) 18,80 mm, (P3) 20,38 mm, (P4) 22,16 mm. Hasil uji DNMRT menunjukkan bahwa rata-rata diameter zona hambat yang terkecil yaitu 14,50 mm pada konsentrasi 10% yang tidak berbeda nyata dengan zona hambat pada perlakuan kontrol dan hand sanitizer daun buta-buta konsentrasi 30%, tetapi berbeda nyata dengan konsentrasi 50% dan 70%. Rata-rata diameter zona hambat yang terbesar yaitu 22,16 mm pada konsentrasi 70% yang berbeda nyata dengan zona hambat pada konsentrasi 10%, tetapi tidak berbeda nyata dengan perlakuan lainnya. Kesimpulan dari penelitian ini adalah hand sanitizer dari ekstrak daun buta-buta berpengaruh terhadap pertumbuhan S. aureus. Konsentrasi yang optimal dari ekstrak daun buta-buta sebagai antibakteri spray hand sanitizer terhadap pertumbuhan S. aureus adalah 50%. This study aims to determine the effect of antibacterial spray hand sanitizer from blind-eyes leaf extract on the growth of S. aureus and to determine the optimal concentration of blind-eyes leaf extract as an antibacterial spray hand sanitizer against the growth of S. aureus bacteria. This research was conducted at the Biology Education Laboratory, Jambi University and UPT Basic and Integrated Laboratory (UPT-LDT) Jambi University from February to March 2022. This study is an experimental study, the research design used was a completely randomized design (CRD) method with 5 treatments and 5 repetitions. The treatments included giving hand sanitizer to S. aureus bacteria, namely control (commercial hand sanitizer), blind leaf extract hand sanitizer with concentrations of 10%, 30%, 50%, and 70%. The research parameters observed included the inhibition zone, pH, and the speed test for hand sanitizer. Data analysis used One Way Analysis of Variance (ANOVA) and if there was an effect of treatment, it was continued with Duncan's New Multiple Range Test (DNMRT) at a 95% confidence level. The results showed that blind-blind leaf extract hand sanitizer had an effect on the growth of S. aureus bacteria with an average control diameter (commercial hand sanitizer) (P0) 19.09 mm, (P1) 14.50 mm, (P2) 18, 80 mm, (P3) 20.38 mm, (P4) 22.16 mm. The results of the DNMRT test showed that the smallest average diameter of the inhibition zone was 14.50 mm at a concentration of 10% which was not significantly different from the inhibition zone in the control treatment and blind leaf hand sanitizer with a concentration of 30%, but significantly different at a concentration of 50%. and 70%. The average diameter of the largest inhibition zone was 22.16 mm at a concentration of 70% which was significantly different from the inhibition zone at a concentration of 10%, but not significantly different from other treatments. The conclusion of this study is that hand sanitizer from blind-eyes leaf extract has an effect on the growth of S. aureus. The optimal concentration of blind-eyes leaf extract as an antibacterial spray hand sanitizer against the growth of S. aureus is 50%.
