Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh 2,4-D terhadap induksi kalus eksplan kopi Komposit Liberika Tungkal dan mendapatkan konsentrasi 2,4-D terbaik untuk induksi kalus eksplan kopi Komposit Liberika Tungkal. Dilaksanakan di Laboratorium Bioteknologi Tanaman, Fakultas Pertanian Universitas Jambi, Desa Mendalo Indah, Kecamatan Jambi luar kota, Kabupaten Muaro Jambi. Penelitian ini dilaksanakan selama lima bulan dari bulan Juni sampai November 2021. Bahan penelitian yang digunakan adalah: Biji kopi liberica, media MS, ZPT 2,4-D, aquades, alkohol, KOH, HCl, bayclin, plastik anti karat, karet band. Alat yang digunakan adalah: timbangan digital, hotplate, autoclave, pH meter, gelas ukur, gelas kimia, cawan petri, botol kultur, batang pengaduk, lemari es, lampu bunsen, korek api, gunting, pinset, pisau bedah, spatula, corong, hand sprayer, label kertas, erlenmeyer, rak kultur, kamera handphone dan alat tulis. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), dengan satu faktor pemberian konsentrasi zat pengatur tumbuh 2,4-D yang terdiri dari 6 taraf perlakuan: 0 mg L-1 (kontrol), 1 mg L-1, 2 mg L-1, 3 mg L-1, 4 mg L-1, 5 mg L-1. Pemberian 2 mg L-1 2,4-D berpengaruh nyata terhadap parameter pengamatan persentase eksplan yang membentuk kalus. Kata kunci: Induksi Kalus, 2,4-D (Dichrolopenoxy Acetic Acid), Kopi Liberika ABSTRACT This study aimed to determine the effect of 2,4-D on callus induction of Liberika Tungkal Composite coffee explants and to obtain the best concentration of 2,4-D for callus induction of Composite Liberika Tungkal coffee explants. Carried out at the Plant Biotechnology Laboratory, Faculty of Agriculture, Jambi University, Mendalo Indah Village, Jambi District outside the city, Muaro Jambi Regency. This research was carried out for five months from June to November 2021. The research materials used were: Liberica coffee seeds, MS media, ZPT 2,4-D, aquades, alcohol, KOH, HCl, bayclin, anti-rust plastic, rubber bands . The tools used are: digital scales, hotplates, autoclaves, pH meters, measuring cups, beakers, petri dishes, culture bottles, stirring rods, refrigerators, Bunsen lamps, matches, scissors, tweezers, scalpels, spatulas, funnels , hand sprayer, label paper, erlenmeyer, culture rack, cellphone camera and stationery. This study used a completely randomized design (CRD), with one factor administration of a concentration of growth regulator 2,4-D consisting of 6 levels of treatment: 0 mg L-1 (control), 1 mg L-1, 2 mg L-1, 3 mg L-1, 4 mg L-1, 5 mg L-1. The administration of 2 mg L-1 2,4-D gave a significant effect on the observation parameters of the percentage of explants that formed callus.
