Abstract

Penelitian ini bertujuan untuk melihat interaksi pada pemberian konsentrasi zat pengatur tumbuh 2,4-D dan NAA terhadap induksi kalus pada eksplan daun kopi liberika Tungkal Jambi (Coffea liberica var. liberica cv. Tungkal Jambi) secara In Vitro dan mendapatkan konsentrasi terbaik dari 2,4-D dan NAA terhadap induksi kalus pada eksplan daun kopi liberika Tungkal Jambi (Coffea liberica var. liberica cv. Tungkal Jambi) secara In Vitro. Penelitian ini dilaksanakan di Laboratorium Bioteknologi Tanaman Fakultas Pertanian, Universitas Jambi, Desa Mendalo Indah, Kecamatan Jambi Luar Kota, Kabupaten Muaro Jambi. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan taraf konsentrasi zat pengatur tumbuh 2,4-D 0 ppm; 1 ppm; 2 ppm; 3 ppm, dan NAA 0 ppm; 2 ppm; 4 ppm; 6 ppm. Setiap perlakuan terdapat 16 perlakuan dan 3 kali ulangan, sehingga didapatkan 48 satuan percobaan. Setiap satuan percobaan terdiri dari 5 botol, sehingga diperoleh 240 botol dengan setiap botol diisi 1 eksplan. Hasil penelitian menunjukkan bahwa pemberian auksin 2,4-D mampu mendapatkan berat kalus terbaik tetapi penambahan NAA dapat memperlambat waktu munculnya kalus. Namun, pemberian konsentrasi 2,4-D dan NAA terhadap induksi kalus daun kopi liberika Tungkal Jambi ini belum mampu memperlihatkan adanya interaksi antara keduanya. This study aimed to see the interaction in the concentration of growth regulators 2,4-D and NAA on callus induction on leaf explants of Tungkal Jambi Liberica coffee (Coffea Liberica var. Liberica cv. Tungkal Jambi) in Vitro and get the best concentration of 2,4-D and NAA on callus induction on leaf explants of Tungkal Jambi Liberica coffee (Coffea Liberica var. Liberica cv. Tungkal Jambi) in Vitro. This research was conducted at the Plant Biotechnology Laboratory, Faculty of Agriculture, Jambi University, Mendalo Indah Village, Jambi Luar Kota District, Muaro Jambi Regency. This study used a completely randomized design (CRD) with concentration levels of growth regulators 2,4-D 0 ppm; 1 ppm; 2 ppm; 3 ppm, and NAA 0 ppm; 2 ppm; 4 ppm; 6 ppm. Each treatment had 16 treatments and 3 replications, resulting in 48 experimental units. Each experimental unit consisted of 5 bottles, resulting in 240 bottles with each bottle filled with 1 explant. The results showed that the provision of auxin 2,4-D was able to get the best callus weight but the addition of NAA could slow down the time of callus emergence. However, the concentration of 2,4-D and NAA on callus induction of Tungkal Jambi Liberika coffee leaves has not been able to show the interaction between the two.
