Abstract

Degenerative diseases such as atherosclerosis, cancer, and premature aging can be caused by oxidative stress. Oxidative stress is triggered by uncontrolled free radical reactions in the body and the inability of endogenous antioxidants to counteract free radicals. Antioxidants are needed to overcome various diseases caused by oxidative stress. Antioxidants can be in the form of synthetic antioxidant compounds and natural antioxidants. One of the plants that can be used as an alternative natural antioxidant is the arecacea family plant. The purpose of this study was to determine and analyze the antioxidant activity and the compounds in the isolate of the red areca root dichloromethane fraction which acts as an antioxidant. The isolates were obtained from the fractionation. For the in vitro antioxidant test using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, 5 series of test concentrations were used with vitamin E as a positive control. The absorbance measurement of the test sample was carried out using a UV-Vis spectrophotometer with a wavelength of 517 nm. Research data were analyzed using one-way ANOVA and LSD and Duncan post hoc follow-up tests. The test results on samples that had been reacted with DPPH after incubation for 30 minutes showed the presence of antioxidant activity which was marked by a change in color from purple to faded purple and then to yellow. Based on the results of the antioxidant test, it can be concluded that the isolate from the dichloromethane fraction has the potential as an antioxidant in counteracting free radicals.
