Abstract
An experiment to study the effect of IAA and BAP on the callus proliferation and on the incidence of vitrification and necrosis of Guichenotia macrantha shoots had been carried out at the Plant Science Laboratory, Victorian College of Agriculture and Horticulture, Melbourne, from January to October 1994. Four levels of IAA (0.00, 0.57, 2.85 and 5.71µM) and four levels of BAP (0.00, 2.22, 4.44 and 8.88µM) were tested in this experiment. The results indicated that either the callus proliferation or the incidence of shoot vitrification and necrosis were significantly affected by the presence of BAP in the culture medium. Callus proliferation was enhanced by BAP at concentration 2.22 and 4.44µM, but was inhibited and concentration 8.88µM. The frequency of shoot vitrification and necrosis increased at BAP concentration 4.44µM or more.
