Abstract

This experiment was conducted at the Plant Tissue Culture Laboratory, Agricultural Faculty Unja, from September until December 1995. Three chemicals tested were Ca-hypochlorite, Na-hypochlorite and HgCl2 which were applied in 4 concentration levels, ie. 0.1, 0.5, 1.0 and 5.0%, and combined with 3 periods of explant dipping, ie. 10, 20 and 30 minutes. In addition to this, the media composition tested were MS, MS½, WPM, B-5, LS and SH. The result indicated that 0.1, 0.5 and 1.0% Ca-hypochlorite combined with 10, 20 and 30 minutes dipping resulted in better effect than 5.0% Ca-hypochlorite combined with 10, 20 and 30 minutes dipping. Furthermore, the 0.1% Na-hypochlorite combined with 30 minutes dipping resulted in better effect than 1.0% Na-hypochlorite+30 minutes dipping and 5.0% Na-hypochlorite+10, 20 and 30 minutes dipping. Among the tested media composition, the basal media MS and WPM produced better effect than MS½, B-5, LS and SH on the percentage of callus formation.
