Abstract

This experiment was conducted at the Plant Tissue Culture Laboratory, Agricultural Faculty Unja, from September until December 1995. Three chemicals tested were Ca-hypochlorite, Na-hypochlorite and HgCl2 which were applied in 4 concentration levels, ie. 0.1, 0.5, 1.0 and 5.0%, and combined with 3 periods of explant dipping, ie. 10, 20 and 30 minutes. In addition to this, the media composition tested were MS, MS½, WPM, B-5, LS and SH. The result indicated that Ca- and Na-hypochlorite were not effective to be used in the sterilisation of single-node citrus explants taken from in vivo culture. Furthermore, even though there was a number of healthy explants, the treatment of HgCl2 concentration combined with dipping period showed a non-significant effect on the culture growth. In addition, there was also no significant effect resulted in six media composition tested.
