Abstract

Guichenotia macrantha Turc.is an Australian native woody ornamental species that has never been propagated via tissue culture technique.For this reason, a preliminary study had been conducted to investigate the sterilization technique and the basal medium composition that is appropriate for this species. The results indicated that the sterilization technique by dipping the G. macrantha seeds in 0.1% hypochlorite solution (Ca- or Na-)resulted in a higher percentage of healthy explants compared to 70% and 40% ethanol, 30% H2O2 or sterile water treatments. Meanwhile, the MS-1/2 medium composition resulted in a higher percentage of seed germination compared to B-5, LS, WPM and MS media. It was recommended to use Ca- or Na-hypochlorite as a sterilization agent, and MS-1/2 as a basal medium for further invertigation.
