Abstract

Penelitian ini bertujuan untuk mengetahui konsentrasi penambahan molasses dalam meningkatkan produksi dan profil gas yang dihasilkan oleh inoculum cairan feses agar dapat menyamai produksi dan profil gas inoculum cairan rumen selama masa inkubasi dengan teknik in vitro. Materi dan peralatan yang digunakan adalah glass syringe, clumper dan declumper, tabung fermentor, water bath, saringan, kain kasa, thermometer, blender, Larutan McDougall, HgCl₂, Satu ekor sapi berpistula rumen, cairan rumen, 300g feses segar, serta bahan untuk komposisi saliva buatan. Rancangan penelitian yang digunakan adalah RAL dengan 5 perlakuan dan 4 ulangan yang terdiri dari RL=cairan rumen, RF=cairan feses, RF2= Cairan feses dan 2 % molases, RF3= Cairan feses dan 3 % molases dan RF4= Cairan feses dan 4 % molases. Peubah yang diamati adalah produksi gas total, potensial produksi gas (b) dan Laju produksi gas (c). Data dianalisis dengan sidik ragam dan bila berpengaruh nyata, maka dilakukan uji lanjut dengan uji BNT. Hasil penelitian menunjukkan bahwa penambahan molasses berpengaruh nyata (P<0,05) terhadap produksi gas total, dimana hasil uji lanjut menunjukkan bahwa perlakuan RL menghasilkan produksi gas total yang berbeda nyata (P<0,05) dengan RF, RF2, RF3 dan RF4. Selanjutnya perlakuan RF juga berbeda nyata (P<0,05) dengan RF2, RF3 dan RF4, perlakuan RF2 berbeda nyata (P<0,05) dengan RF3 dan RF4, sedangkan RF3 tidak berbeda (P>0,05) dengan RF4. Selanjutnya penambahan molasses berpengaruh nyata (P<0,05) terhadap potensial produksi gas ( b ), dimana perlakuan RL berbeda nyata (P<0,05) dengan RF, RF2, RF3 dan RF4, sedangkan perlakuan RF tidak berbeda (P>0,05) dengan RF2, tetapi berbeda nyata (P<0,05) dengan RF3 dan RF4. Sementara itu, perlakuan RF3 tidak berbeda (P>0,05) dengan RF4. Penambahan molasses juga berpengaruh nyata (P<0,05) terhadap Laju produksi gas ( c ), dimana perlakuan RL tidak berbeda nyata (P>0,05) dengan RF, tetapi berbeda nyata (P<0,05) dengan RF2, RF3 dan RF4. Selanjutnya RF berbeda nyata (P<0,05) dengan RF2, RF3 dan RF4. Perlakuan RF2 berbeda nyata (P<0,05) dengan RF3 dan RF4, sedangkan RF3 tidak berbeda (P>0,05) dengan RF4.. Kesimpulan konsentrasi penambahan molasses sangat berpengaruh pada produksi gas dan profil gas yang dihasilkan inoculum cairan feses namun belum dapat menyamai produksi gas dan profil gas yang dihasilkan oleh inoculum cairan rumen selama masa inkubasi dengan teknik in vitro. This study aims to determine the concentration of molasses addition in increasing the production and gas profile produced by fecal fluid inoculum in order to match the production and gas profile of rumen fluid inoculum during the incubation period using in vitro techniques. The materials and equipment used were glass syringe, clumper and declumper, fermentor tube, water bath, filter, gauze, thermometer, blender, McDougall's solution, HgCl₂, one cow with rumen fistula, rumen fluid, 300g of fresh feces, and materials for artificial saliva composition. The research design used was RAL with 5 treatments and 4 replications consisting of RL = rumen fluid, RF = feces fluid, RF2 = Feces fluid and 2% molasses, RF3 = Feces fluid and 3% molasses and RF4 = Feces fluid and 4% molasses. The variables observed were total gas production, gas production potential (b) and gas production rate (c). Data were analyzed by analysis of variance and if there was a significant effect, further testing was carried out using the BNT test. The results showed that the addition of molasses had a significant effect (P<0.05) on total gas production, where the results of further testing showed that the RL treatment produced a total gas production that was significantly different (P<0.05) from RF, RF2, RF3 and RF4. Furthermore, the RF treatment was also significantly different (P<0.05) from RF2, RF3 and RF4, the RF2 treatment was significantly different (P<0.05) from RF3 and RF4, while RF3 was not different (P>0.05) from RF4. Furthermore, the addition of molasses had a significant effect (P<0.05) on the potential gas production (b), where the RL treatment was significantly different (P<0.05) from RF, RF2, RF3 and RF4, while the RF treatment was not different (P>0.05) from RF2, but significantly different (P<0.05) from RF3 and RF4. Meanwhile, the RF3 treatment was not different (P>0.05) from RF4. The addition of molasses also had a significant effect (P<0.05) on the gas production rate (c), where the RL treatment was not significantly different (P>0.05) from RF, but significantly different (P<0.05) from RF2, RF3 and RF4. Furthermore, RF was significantly different (P<0.05) from RF2, RF3 and RF4. The RF2 treatment was significantly different (P<0.05) from RF3 and RF4, while RF3 was not different (P>0.05) from RF4. The conclusion is that the concentration of molasses addition greatly affects gas production and the gas profile produced by the fecal fluid inoculum but has not been able to match the gas production and gas profile produced by the rumen fluid inoculum during the incubation period using the in vitro technique.
