Abstract

Latar belakang: MC4R merupakan salah satu gen yang berfungsi dalam mengatur pola makan seseorang. Terjadinya mutasi pada gen MC4R dengan SNP rs17782313 mengakibatkan hiperfagia, peningkatan berat badan, dan obesitas sentral. Obesitas sentral merupakan kejadian di mana terdapat penumpukan lemak di rongga abdomen dan dapat diukur besarnya menggunakan waist-hip ratio (WHR). Penelitian ini bertujuan untuk menganalisis hubungan polimorfisme gen MC4R rs17782313 terhadap WHR pada populasi di Kota Jambi. Metode penelitian: Desain penelitian ini adalah unmatched case-control, melibatkan 106 sampel DNA dari pasien dengan WHR meningkat dan normal dengan jumlah sampel masing-masing adalah 53 dan dengan rentang usia 18-54 tahun. Sampel dibedakan berdasarkan hasil WHR dan dikatakan meningkat jika ≥0,90 pada laki-laki dan ≥85 pada perempuan. Genotyping dilakukan menggunakan teknik Tetra ARMS-PCR. Analisis statistik yang digunakan pada penelitian ini adalah Chi-square, Mann-Whitney U, dan Independent Samples t-Test. Hasil: Hasil statistik menunjukkan hasil sebagai berikut: genotipe TC model aditif (OR 2,932; CI 1,136-7,568; p=0,023), genotipe homozigot CC (OR 1,303; CI 0,174-9,742; p=1,000), dan genotipe model resesif CC+TC (OR 2,606; CI 1,076-6,310; p=0,031). Kesimpulan: Polimorfisme gen MC4R rs17782313 genotipe TC model aditif dan genotipe model resesif CC+TC memiliki hubungan yang signifikan terhadap WHR pada populasi ini. Kata kunci: MC4R, Obesitas Sentral, Polimorfisme Gen, WHR Background: MC4R is one of the genes that functions in regulating a person's diet. The occurrence of mutations in the MC4R gene with SNP rs17782313 results in hyperphagia, weight gain, and central obesity. Central obesity is an event in which there is a buildup of fat in the abdominal cavity and can be measured using waist-hip ratio (WHR). This study aims to analyze the relationship of MC4R gene polymorphism rs17782313 to WHR in the population in Jambi City. Methods: The design of this study was unmatched case-control, involving 106 DNA samples from patients with increased and normal WHR with a sample count of 53 and with an age range of 18-54 years. The sample was differentiated based on the WHR results and was said to increase if ≥0.90 in males and ≥85 in females. Genotyping was carried out using the Tetra ARMS-PCR technique. The statistical analysis used in this study was Chi-square, Mann-Whitney U, and Independent Samples t-Test. Results: Statistical results show the following results: additive model TC genotype (OR 2,932; CI 1,136-7,568; p=0.023), homozygous CC genotype (OR 1.303; CI 0.174-9.742; p=1,000), and recessive model genotype CC+TC (OR 2,606; CI 1,076-6,310; p=0.031). Conclusion: The polymorphism of the MC4R gene rs17782313 of the additive model TC genotype and the CC+TC recessive model genotype had a significant relationship with WHR in this population.
