Abstract

RINGKASAN Kayu secang (Caesalpinia sappan L.) dapat digunakan untuk kesehatan dan pengobatan berbagai penyakit. Secang mengandung senyawa kimia yang dapat digunakan sebagai antibakteri. Senyawa antibakteri pada kayu secang adalah senyawa brazilin, flavonoid, alkaloid, tanin, saponin dan terpenoid. Salah satu bakteri penyebab penyakit pada ikan adalah Aeromonas caviae khususnya menyerang ikan air tawar. A. caviae mampu menyebabkan penyakit Motile Aeromonas Septicemia dengan gejala klinis luka kemerahan pada bagian sungut, sirip dan ekor, pendarahan pada tubuh ikan yang akan berubah menjadi borok (hemorrhagic) pada bagian kulit, kemampuan berenang ikan menurun dan sering berada diatas permukaan air, serta pendarahan pada organ hati, ginjal dan limfa. Tujuan penelitian ini adalah untuk melihat potensi ekstrak kayu secang terhadap pertumbuhan A. caviae, dan untuk mengetahui Konsentrasi Hambat Minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) ekstrak kayu secang terhadap A. caviae. Penelitian ini bersifat eksperimental yang dilakukan di Laboratorium Bioteknologi dan Tanaman obat dan di Laboratorium Fakultas Sains dan Teknologi Universitas Jambi. Penelitian dilakukan dengan beberapa tahapan yaitu maserasi dan esktraksi sampel, peremajaan bakteri, pengukuran kurva pertumbuhan bakteri dan uji potensi antibakteri ekstrak kayu secang dengan konsentrasi 1,25%, 2,5%, 5%, 10%, dan 20% terhadap A. caviae. Dari hasil penelitian didapatkan hasil bahwa ekstrak kayu secang memiliki potensi antibakteri terhadap A. caviae karena terjadinya penurunan nilai absorbansi setelah inkubasi. Berdasarkan hasil pengukuran absorbansi menggunakan spektrofotometer UV-vis 600 nm, konsentrasi terkecil yang mengalami penurunan nilai absorbansi adalah konsentrasi 2,5% sehingga Konsentrasi Hambat Minimum (KHM) ekstrak kayu secang terhadap A. caviae yaitu konsentrasi 2,5% dan konsentrasi terkecil yang mampu membunuh pertumbuhan A. caviae adalah konsentrasi 5% sehingga Konsentrasi Bunuh Minimum (KBM) ekstrak kayu secang terhadap A. caviae yaitu konsentrasi 5%. SUMMARY Sappan wood (Caesalpinia sappan L.) can be used for health and treatment of various diseases. Sappan contains chemical compounds that can be used as antibacterial. Antibacterial compounds found in sappan wood are brazilin, flavonoids, alkaloids, tannins, saponins and terpenoids. One of the bacteria that causes disease in fish is Aeromonas caviae, especially attacking freshwater fish. A. caviae can cause Motile Aeromonas Septicemia disease with clinical symptoms in the form of reddish wounds on the barbels, fins and tail, bleeding in the fish's body which will turn into ulcers (hemorrhagic) on the skin, decreased swimming ability of fish and often above the water surface, and bleeding in the liver, kidneys and spleen. The purpose of this study was to see the potential of sappan wood extract on the growth of A. caviae, and to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bacterial Concentration (MBC) of sappan wood extract on A. caviae. This research is experimental in nature conducted in the Laboratory of Biotechnology and Plant Medicine and in the Laboratory of the Faculty of Science and Technology, University of Jambi. The research was conducted in several stages, namely maceration and sample extraction, bacterial rejuvenation, measurement of bacterial growth curves and antibacterial potential testing of sappanwood extract with concentrations of 1.25%, 2.5%, 5%, 10%, and 20% against A. caviae. The results showed that sappan wood extract has antibacterial potential against A. caviae due to a decrease in absorbance values after incubation. Based on the results of absorbance measurements using a 600 nm UV-vis spectrophotometer, the smallest concentration that experienced a decrease in absorbance value was a concentration of 2.5% so that the Minimum Inhibitory Concentration (MIC) of sappan wood extract against A. caviae was a concentration of 2.5% and the smallest concentration that could kill the growth of A. caviae was a concentration of 5% so that the Minimum Killing Concentration (MBC) of sappan wood extract against A. caviae was a concentration of 5%.
