Abstract

RINGKASAN Kayu secang (Caesalpinia sappan L.) telah dilaporkan memiliki berbagai senyawa metabolit, diantaranya alkaloid, flavonoid, saponin, tanin dan terpenoid. Kayu secang memiliki senyawa identitas berupa brazilin. Karena senyawa metabolit yang terdapat didalam kayu secang, maka kayu secang memiliki potensi antibakteri. Jenis bakteri penyebab infeksi pada budidaya air tawar ialah Aeromonas sobria. Bakteri ini menyebabkan infeksi Motil Aeromonas Septicemia (MAS) pada budidaya perikanan air tawar. Penanganan infeksi ini umumnya menggunakan obat obatan berupa antibiotik. Namun penggunaan antibiotik secara berkepanjangan dapat menyebabkan resistensi bakteri terhadap antibiotik. Galur bakteri resisten antibiotik menyebabkan pengobatan menjadi tidak efisien bahkan residu antibiotik tersebut dapat memiliki potensi untuk tersalurkan kepada manusia melalui rantai makanan. Selain itu, telah dilaporkan dari berbagai sumber terkait resistensi A. sobria terhadap antibiotik. Oleh karena itu diperlukan alternatif pengobatan yang lebih aman seperti penggunaan tumbuhan dengan memanfaatkan metabolit sekunder tumbuhan tersebut. Tujuan dari penelitian ini untuk mengetahui potensi ekstrak kayu secang sebagai antibakteri terhadap pertumbuhan A.sobria serta untuk mengetahui Konsentrasi Hambat Minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM) ekstrak kayu secang terhadap A. sobria. Penelitian dilaksanakan di Laboratorium Bioteknologi dan Tanaman Obat Fakultas Sains dan Teknologi serta Laboratorium Pengujian Fakultas Sains dan Teknologi. Penelitian ini dilakukan dengan berberapa tahapan diantaranya persiapan sampel, maserasi, evaporasi, pembuatan konsentrasi ekstrak dan kontrol, pengukuran kurva pertumbuhan, uji potensi antibakteri dengan metode dilusi yang terdiri dari uji KHM dan uji KBM. Dari penelitian yang telah dilakukan didapatkan hasil bahwa ekstrak kayu secang dapat menghambat dan juga membunuh bakteri Aeromonas sobria pada konsentrasi 10%. Kata kunci: Kayu secang (Caesalpinia sappan L.), Aeromonas sobria, Potensi antibakteri, Konsentrasi Hambat Minimum (KHM), Konsentrasi Bunuh Minimum (KBM) SUMMARY Sappan wood (Caesalpinia sappan L.) has been reported to have various metabolite compounds, including alkaloids, flavonoids, saponins, tannins and terpenoids. Sappan wood has an identity compound in the form of brazilin. Because of the metabolite compounds contained in sappan wood, sappan wood has antibacterial potential. The type of bacteria that causes infections in freshwater aquaculture is Aeromonas sobria. This bacteria causes Motil Aeromonas Septicemia (MAS) infection in freshwater fisheries. Treatment of this infection generally uses drugs in the form of antibiotics. However, prolonged use of antibiotics can cause bacterial resistance to antibiotics. Antibiotic-resistant bacterial strains make treatment inefficient and even antibiotic residues can have the potential to be transmitted to humans through the food chain. In addition, it has been reported from various sources regarding A. sobria resistance to antibiotics. Therefore, safer alternative treatments are needed, such as the use of plants by utilizing the secondary metabolites of these plants.The purpose of this study was to determine the potential of sappanwood extract as an antibacterial against the growth of A. sobria and to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of sappanwood extract against A. sobria. The study was conducted in the Biotechnology and Medicinal Plants Laboratory of the Faculty of Science and Technology and the Testing Laboratory of the Faculty of Science and Technology. This study was conducted in several stages including sample preparation, maceration, evaporation, making extract concentrations and controls, measuring growth curves, testing antibacterial potential using the dilution method consisting of the MIC test and the MBC test. From the research that has been carried out, it was found that sappanwood extract can inhibit and also kill Aeromonas sobria bacteria at a concentration of 10%.
