Abstract
RINGKASAN Bioremediasi adalah metode pembersihan lingkungan yang digunakan untuk mengurangi pencemaran organik maupun anorganik dengan bantuan agen biologi seperti bakteri. Bioremediasi bertujuan untuk mengurangi konsentrasi pencemar atau zat pencemar berbahaya ke level yang tidak berbahaya melalui proses degradasi, detoksifikasi, mineralisasi, dan transformasi. Bakteri hidrokarbonoklastik merupakan salah satu bakteri yang memiliki kemampuan untuk mendegradasi minyak solar. Penelitian terdahulu telah berhasil mengidentifikasi bakteri indigenous pendegradasi hidrokarbon dari Pelabuhan Pelabuhan Kuala Tungkal Ilir dan Roro Kuala Tungkal Kabupaten Tanjung Jabung Barat, dilaporkan bahwa terdapat 20 isolat bakteri yang memiliki kemampuan dalam mendegradasi senyawa hidrokarbon. Tujuan dari penelitian ini yaitu menguji apakah isolat bakteri potensial pendegradasi minyak solar mampu menurunkan nilai Total Petroleum Hydrocarbon (TPH) dan seberapa besar kemampuan isolat bakteri potensial pendegradasi minyak solar dalam menurunkan nilai TPH. Penelitian ini bersifat kuantitatif eksperimental yang dilakukan di Laboratorium Agroindustri Tanaman Obat dan Bioteknologi serta Green House Biologi Fakultas Sains dan Teknologi Universitas Jambi. Metode yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 6 kali pengulangan. Tahap penelitian yang dimulai dari sterilisasi alat dan bahan, pembuatan media uji, uji pendahuluan, peremajaan bakteri pendegradasi, pembuatan suspensi bakteri, uji kemampuan isolat bakteri potensial pendegradasi minyak solar, dan pengukuran TPH dengan metode gravimetri. Dalam penelitian ini terdapat uji pendahuluan yang berfungsi untuk menyeleksi ke 20 isolat bakteri dari penelitian terdahulu untuk nantinya diambil 1 isolat bakteri yang paling berpotensi dalam mendegradasi minyak. Terdapat 1 isolat bakteri yang paling berpotensi yaitu isolat bakteri dengan kode RT4. Hasil seleksi mendapatkan 1 isolat potensial yang diuji pada penelitian ini. Penelitian ini menggunakan isolat bekteri dengan konsentrasi 10% dan dengan konsetrasi variasi minyak solar yang digunakan dalam uji sebanyak 5%, 10%, dan 15%. Hasil dari pengukuran TPH dengan metode gravimetri menunjukkan bahwa isolat bakteri potensial memiliki kemampuan dalam menurunkan nilai TPH. Analisis data menggunakan ANOVA (Analisis Of Variance) dan dilanjutkan dengan menggunakan uji Post Hoc Tukey. Berdasarkan hasil uji lanjut didapatkan kemampuan terbaik bakteri dalam mendegradasi terjadi di konsentrasi minyak solar 15%. Dari penelitian ini, diharapkan bahwa isolat bakteri dengan kode isolat RT4 dapat memberikan kontribusi signifikan dalam upaya bioremediasi di lingkungan perairan laut. Bioremediation is an environmental clean-up method used to reduce organic and inorganic pollution with the help of biological agents such as bacteria. Bioremediation aims to reduce the concentration of pollutants or hazardous contaminants to harmless levels through the processes of degradation, detoxification, mineralisation, and transformation. Hydrocarbonoclastic bacteria is one of the bacteria that has the ability to degrade diesel oil. Previous research has successfully identified indigenous bacteria degrading hydrocarbons from Kuala Tungkal Ilir Port and Kuala Tungkal Roro Port, West Tanjung Jabung Regency, reported that there are 20 bacterial isolates that have the ability to degrade hydrocarbon compounds. The purpose of this study is to test whether potential bacterial isolates of diesel oil degraders are able to reduce the value of Total Petroleum Hydrocarbon (TPH) and how much the ability of potential bacterial isolates of diesel oil degraders in reducing the value of TPH. This research is a quantitative experimental conducted at the Laboratory of Medicinal Plant Agroindustry and Biotechnology and Green House Biology, Faculty of Science and Technology, Jambi University. The method used in this study was a completely randomised design (CRD) with 4 treatments and 6 repetitions. The research stage starts from sterilisation of tools and materials, making test media, preliminary tests, rejuvenation of degrading bacteria, making bacterial suspensions, testing the ability of potential bacterial isolates to degrade diesel oil, and measuring TPH by gravimetric method. In this study, there is a preliminary test that serves to select 20 bacterial isolates from previous studies to later take 1 bacterial isolate that has the most potential in degrading oil. There is 1 bacterial isolate that has the most potential, namely the bacterial isolate with the code RT4. The selection results get 1 potential isolate tested in this study. This study used bacterial isolates with a concentration of 10% and with the concentration of diesel oil variations used in the test as much as 5%, 10%, and 15%. The results of TPH measurement by gravimetric method showed that potential bacterial isolates have the ability to reduce TPH value. Data analysis was performed using ANOVA (Analysis of Variance) and followed by Tukey's Post Hoc test. Based on the results of the follow-up test, the best ability of bacteria to degrade occurred at a diesel oil concentration of 15%. From this study, it is expected that bacterial isolates with isolate code RT4 can make a significant contribution to bioremediation efforts in the marine environment
